I wouldn't bother.  Substituting an uninsulated ceiling/floor for an 
insulated one (plus some walls) is not much of an improvement and can 
cause lots of problems, for instance, with condensation.

You want the simplest arrangement in terms of air flow.  That would 
recommend putting the registers near (directly above) the woodstove, 
and having a place where the cold air can flow down somewhere else.  If 
the registers are going to be in a bedroom, I would put in a small 
dogleg to maintain a bit of privacy.
